The Phragmoplast

Pflanzenzellen sind von einer starren Zellwand umgeben, die es den Zellen erschwert, sich durch Zusammenziehen von außen zu teilen. Pflanzenzellen teilen sich also, indem sie eine neue Zellwand zwischen den beiden Tochterkernen "von innen nach außen" bilden. Eine spezialisierte Struktur in Pflanzen, die Phragmoplasten, steuert die Bildung neuer Zellwände.
Die Zellteilungsebene, die der zukünftige Ort der Zellwandbildung ist, ist durch eine Präprophase-Bande aus Mikrotubuli und Aktinfilamenten gekennzeichnet, die sich während der G1-Phase des Zellzyklus bildet. Die Präprophasenbande baut sich an der Zellrinde an und verschwindet zu Beginn der M-Phase.
Während der Mitose trennen sich die Chromosomen und beginnen, sich zu entgegengesetzten Polen zu bewegen. An den Polen bildet sich die Kernhülle um die getrennten Chromosomen und die Spindeln beginnen zu verschwinden. Die restlichen Mikrotubuli der mitotischen Spindel bilden den Phragmoplasten.
Die Mikrotubuli der Phragmoplasten sind polar, mit Plus-Enden in der Nähe der Äquatorebene und Minus-Enden in der Nähe der Pole.
Golgi-Körper sezernieren kleine Vesikel, die von Motorproteinen entlang der Mikrotubuli zum Zellzentrum transportiert werden. Diese Vesikel sind mit Polysacchariden und Glykoproteinen gefüllt, zwei wichtigen Komponenten, die für die Synthese neuer Zellwände benötigt werden. Durch die Verschmelzung von Vesikeln entsteht eine scheibenförmige Struktur, die als frühe Zellplatte bezeichnet wird.
Die Mikrotubuli des Phragmoplasten werden erfolgreich an den freien Rändern der Zellplatte regeneriert, so dass der Phragmoplast die Platte lateral verlängern kann. Die neue Zellplatte verbindet sich mit den Mutterzellwänden an der Stelle, die vor der M-Phase durch die Präprophasenbande markiert war. Die Plasmamembran verschmilzt mit der Zellplatte und es bildet sich eine neue Zellwand, die die beiden Tochterzellen trennt.

der phragmoplast

Die Zellteilung ist essentiell für das Wachstum und die Entwicklung von Organismen. In tierischen Zellen bildet der zentrale Spindelapparat zusammen mit seinen assoziierten Proteinen den Midbody, eine Struktur, die eine wesentliche Rolle bei der Zytokinese spielt. In Pflanzenzellen reift der zentrale Spindelapparat zusammen mit den Mikrotubuli, Aktin und anderen Zellbestandteilen zum Phragmoplasten heran, der für die Zytokinese notwendig ist. Im Gegensatz zum stationären Midbody dehnt sich der Phragmoplast zentrifugal aus, was schließlich zur Bildung der neuen Zellwand führt.

Der ausgereifte Phragmoplast ist donutförmig und enthält eine äußere Führungsregion, eine mittlere Übergangszone und eine innere Nachhinkregion. In der Führungsregion werden kontinuierlich neue Mikrotubuli zusammengesetzt. Die Nukleation der Mikrotubuli wird durch den γ-Tubulin-Ringkomplex (γ-TuRC) eingeleitet, der sechs Untereinheiten—GCP1 bis GCP6—enthält. Allerdings kann γ-TuRC nicht direkt mit den Mikrotubuli interagieren; daher vermittelt Augmin, ein mit Mikrotubuli interagierender Proteinkomplex, die Rekrutierung von γ-TuRC zu den Mikrotubuli. Die Mikrotubuli im Phragmoplasten dienen als Bahnen für den Transport der Sekretvesikel, die vom nahegelegenen Golgi-Apparat produziert werden.

In der mittleren Übergangszone wandeln sich die Vesikel zunächst in röhrenähnliche Strukturen um und vernetzen sich anschließend zu einem ineinander verwobenen tubulären Netzwerk. Die kontinuierliche Hinzufügung von Sekretvesikeln führt zum Auswachsen des tubulären Netzwerks, bis es auf die Membran der Mutterzelle trifft. Die Vesikelmembranen werden verwendet, um die Plasmamembran der Tochterzellen zu bilden, während die Biomoleküle in den Vesikeln verwendet werden, um die Zellplatte zwischen den Membranen der beiden Tochterzellen zu bilden.

In der Nachhinkregion depolymerisieren die Mikrotubuli, wobei der ausgereifte Teil der Zellplatte zurückbleibt. Die vollständig gebildete Zellplatte entwickelt sich anschließend zur Zellwand, die die beiden Tochterzellen trennt. Die Polymerisation der Mikrotubuli am führenden Ende und die Depolymerisation der Mikrotubuli in der Nachhinkregion bewirken die zentrifugale Ausdehnung des Phragmoplasten und der Zellplatte, bis letztere mit der Zellwand der Elternzelle zusammenwächst. Ein Versagen der Phragmoplastfunktionen kann zu multinukleierten Zellen und embryonal letalen Phänotypen führen.

Plant cells build a new wall during cytokinesis with help from the phragmoplast. This plant structure forms from the central spindle and related cell parts as the cell finishes dividing. In animal cells, the comparable structure is the midbody, but in plants the phragmoplast expands outward to guide new wall formation.

The mature phragmoplast has a donut-like shape. It has an outer leading region, a middle transition zone, and an inner lagging region. New microtubules are assembled continuously in the leading region. Microtubule nucleation begins with the gamma-tubulin ring complex, or gamma-TuRC, which contains six subunits called GCP1 through GCP6. Because gamma-TuRC cannot attach to microtubules on its own, augmin, a microtubule-interacting protein complex, helps recruit it to the microtubules.

The microtubules in the phragmoplast act as tracks for secretory vesicles from the nearby Golgi body. In the middle transition zone, these vesicles first change into tubule-like structures. They then connect to form an intertwined tubular network. As more vesicles are added, the network grows outward until it reaches the parental cell membrane.

The vesicle membranes become the plasma membrane of the daughter cells. The materials inside the vesicles help build the cell plate between the two daughter-cell membranes. In the lagging region, the microtubules break down, leaving the mature part of the cell plate behind. The finished cell plate then develops into the cell wall that separates the two daughter cells.

This outward growth depends on microtubules being built at the leading end and broken down at the lagging end. If phragmoplast function fails, cells can become multinucleated. It can also lead to embryonic lethal phenotypes.
