Distribution of Cytoplasmic Content

Das Zytoplasma einer Zelle enthält viele verschiedene Organellen. Jede Organelle unterscheidet sich in ihrer Struktur, Funktion und Kopienzahl; Daher muss eine Zelle unterschiedliche Strategien anwenden, um jeden Organellentyp richtig in seine Tochterzellen zu unterteilen.
Die Kernhülle und das endoplasmatische Retikulum sind direkt miteinander verbunden, und daher ist ihre Verteilung an die Tochterzellen gekoppelt.
In der Prophase verschmelzen die Kernhülle und die zugehörigen Proteine mit dem endoplasmatischen Retikulum, wobei sich das blattförmige endoplasmatische Retikulum in eine röhrenförmige Struktur verwandelt und sich während der Metaphase in der Nähe der Zellrinde neu organisiert.
Nach der Chromosomensegregation umgibt das endoplasmatische Retikulum die Schwesterchromatiden und reformiert die Kernhülle.
Bei der Mitose werden die Mitochondrien gespalten, um fragmentierte Mitochondrien zu produzieren, die sich in der gesamten Zelle ausbreiten.
Es wird angenommen, dass diese fragmentierten Mitochondrien während der Zellteilung durch einen stochastischen oder zufälligen Prozess an die Tochterzellen verteilt werden und dann fusionieren, um ihre Form und Größe wiederzuerlangen.
Eine einzige Kopie des Golgi-Körpers ist in der Zelle vorhanden, und ihre gleichmäßige Verteilung ist für das Überleben der Zellen von entscheidender Bedeutung. Der Golgi-Körper zerlegt sich schrittweise, bevor er sich in die Tochterzellen aufteilt.
Das Golgi-Band mit den gestapelten Zisternen zerfällt in mehrere Mini-Stapel, die dann weiter in Vesikel zerlegt werden.
In der Anaphase verschmelzen die Golgi-Vesikel zu Zisternen, die dann wieder gestapelt werden. Dann bilden sich in der Telophase kleine und große Zwillings-Golgis in der Nähe des Mittelkörpers bzw. des Zentrosoms.
Während der Zytokinese verschmelzen schließlich die kleineren Zwillings-Golgi-Körper mit den größeren und bilden in jeder Tochterzelle den einzelnen Golgi-Körper.
Eine symmetrische Teilung findet in den meisten Zellen statt, bei denen eine Zelle zwei identische Tochterzellen mit dem gleichen Schicksal hervorbringt.
In einigen Zellen kommt es zu einer asymmetrischen Teilung, bei der die Determinanten des Zellschicksals ungleich vererbt werden, was zur Produktion nicht identischer Tochterzellen führt.
Zum Beispiel können Stammzellen eine asymmetrische Zellteilung durchlaufen, um eine Tochterzelle zu produzieren, die sich wie die Elternstammzelle verhält, während die andere Tochterzelle einer anderen Zelllinie folgen kann.

Organellen teilen sich und wachsen vor der Zellteilung, können aber nicht de novo synthetisiert werden; Daher müssen Zellen mindestens eine Kopie jeder Organelle erhalten, um zu überleben. Derzeit sind viele Einzelheiten der Verteilung der Organellen noch nicht vollständig geklärt.

Verteilung zytoplasmatischer Determinanten

Das Zytoplasma enthält verschiedene Organellen sowie Salze, Proteine und Wasser. Die Verteilung kleiner Organellen wie Peroxisomen und Lysosomen kann durch einfache Diffusion erklärt werden. Da jede Zelle über mehrere Kopien jeder Organelle verfügt, besteht eine hohe Wahrscheinlichkeit, dass Tochterzellen mindestens eine Kopie der Organelle erhalten. Allerdings erfordert die Verteilung großer Organellen wie des ER oder des Chloroplasten komplexere, evolutionär konservierte Mechanismen.

Jüngste Fortschritte bei der Verbreitung von Golgi-Körpern

Der Golgi-Apparat bricht zusammen, wenn die Zellen in die Mitose eintreten. Zwei mögliche Modelle könnten die Verteilung des Golgi-Apparats in Tochterzellen erklären. Nach dem ersten Modell werden während der Prophase die Golgi-Tubuli gespalten und kleine Golgi-Stapel freigesetzt. Diese sind weiter in Vesikel und Tubuli fragmentiert. Diese mitotischen Golgi-Cluster (MGC) verteilen sich im Zytoplasma und segregieren während der Anaphase in die sich teilenden Zellen. Spindelmikrotubuli steuern die Verteilung von MGC auf geordnete Weise. Das zweite Modell geht davon aus, dass die Golgi-Proteine und -Membranen zu Beginn der Mitose im ER resorbiert werden, was es dem Golgi ermöglicht, sich mit dem ER zu teilen.

Mitochondrien sind entscheidende Organellen für das Überleben und Wachstum der Zellen. Während der Mitose werden Mitochondrien in kleinere Fragmente geteilt, die sich dann zwischen den Tochterzellen aufteilen können. Ein konserviertes Protein DRP1 verengt die äußere Mitochondrienmembran und führt zu einer Mitochondrienspaltung. Cyclin B-Cdk1 und Aurora-A, zwei mitotische Kinasen, regulieren die Aktivierung von DRP1.

Das Interphase-Endoplasmatische Retikulum (ER) ist eine kontinuierliche Membran, die die Membranen für Golgi-Körper, Endolysosomen, Vesikel und die Plasmamembran synthetisiert. Während der Mitose kann es zu einer Fragmentierung des ER kommen oder es kann als Zisterne verbleiben. In HeLa-Zellen wird das ER in Form von Zisternen aufrechterhalten und ist auf die Zellrinde beschränkt. Das ER ist ungleichmäßig verteilt und die zentralen Zisternen sind mit der Spindel verbunden. Cytokinesis splits a cell’s chromosomes and organelles into two daughter cells. Before cell division, organelles grow and divide. They cannot be made from scratch, so each daughter cell must inherit at least one copy to survive. Many details of this organelle sorting process are still not fully known.

The cytoplasm contains organelles, salts, proteins, and water. Small organelles such as peroxisomes and lysosomes can be shared by simple diffusion. Because cells contain many copies of these organelles, each daughter cell has a high chance of getting at least one. Larger organelles, such as the endoplasmic reticulum (ER) and chloroplasts, need more complex and conserved mechanisms.

The Golgi apparatus shows one of the best-studied cases. As cells enter mitosis, the Golgi breaks down. One model says the Golgi tubules are cut during prophase, and small stacks are released. These stacks are broken into vesicles and tubules, forming mitotic Golgi clusters, or MGCs, that spread through the cytoplasm and divide during anaphase with help from spindle microtubules. A second model suggests that Golgi proteins and membranes are taken back into the ER at the start of mitosis, so the Golgi is partitioned with the ER.

Mitochondria also change form before they are passed to daughter cells. These organelles are important for cell survival and growth. During mitosis, mitochondria break into smaller pieces by fission, which is the splitting of one organelle into two. The protein DRP1 constricts the outer mitochondrial membrane and drives this process. Two mitotic kinases, Cyclin B-Cdk1 and Aurora-A, regulate DRP1 activity.

The ER can also follow different patterns during mitosis. In interphase, the ER is a continuous membrane that helps make membranes for Golgi bodies, endolysosomes, vesicles, and the plasma membrane. During mitosis, it may fragment or stay as cisternae, which are flattened membrane sacs. In HeLa cells, the ER stays as cisternae and is kept near the cell cortex. The ER is not spread evenly, and the cisternae in the center are linked to the spindle. The ER may be tethered to the cortex wall before it is sorted into the daughter cells.
