Allosteric Proteins-ATCase

Proteine, die verknüpfte Bindungsstellen enthalten, werden als allosterische Proteine bezeichnet und können von verschiedenen Typen sein, wie z. B. Enzyme, Rezeptoren, Strukturproteine und Motorproteine. 
Zum Beispiel ist Aspartat-Transcarbamoylase, auch bekannt als ATCase, ein großes Enzym, das sechs katalytische Untereinheiten enthält, von denen jede eine Substratbindungsstelle hat. Es hat auch sechs zusätzliche regulatorische Untereinheiten, jede mit einer Stelle, die sowohl Pyrimidine als auch Purine, die Bausteine von DNA und RNA, binden kann.

Bindungsstellenverknüpfungen können die Funktion eines Proteins regulieren. Beispielsweise wird die Enzymaktivität oft durch einen Rückkopplungsmechanismus reguliert, bei dem das Endprodukt des biochemischen Prozesses als Hemmstoff dient.

Aspartat-Transcarbamoylase (ATCase) ist ein zytosolisches Enzym, das die Kondensation von L-Aspartat und Carbamoylphosphat zu N-Carbamoyl-L-Aspartat katalysiert. Diese Reaktion ist der erste Schritt in der Pyrimidinbiosynthese. UTP und CTP, die Endprodukte des Pyrimidinsynthesewegs, hemmen die Aktivität der ATCase, des Enzyms, das den ersten wesentlichen Schritt dieses Weges katalysiert. Die Bindung von UTP und CTP an das Enzym reguliert den verbundenen katalytischen Ort negativ, wenn die Konzentration der Pyrimidine im Vergleich zur Konzentration der Purine in der Zelle hoch ist. Dieses Phänomen wird als Feedback-Inhibition bezeichnet und ist entscheidend für die Aufrechterhaltung der richtigen Mengen an Metaboliten in einem Organismus.

ATCase ist Teil des CAD-Multienzymkomplexes, der zusammen mit Carbamoylphosphat-Synthetase II und Dihydroorotase zum Pyrimidinbiosyntheseweg gehört. Pyrimidine sind essentiell für die DNA-Synthese während der Zellteilung, daher kann die Hemmung der ATCase-Aktivität das Tumorwachstum bei Krebs verlangsamen.

ATCase is a key enzyme in pyrimidine synthesis. It is a cytosolic enzyme that catalyzes the condensation of L-aspartate and carbamoyl phosphate to form N-carbamoyl-L-aspartate. This reaction starts the pathway that makes pyrimidines.

ATCase is regulated by feedback inhibition. The end products UTP and CTP bind to the enzyme and reduce its activity when pyrimidine levels are high compared with purines in the cell. This binding at one site affects the linked catalytic site, which helps keep metabolite levels balanced.

ATCase also works as part of the CAD multi-enzyme complex. CAD includes carbamoyl phosphate synthetase II and dihydroorotase in the pyrimidine biosynthesis pathway. Because pyrimidines are needed for DNA synthesis during cell division, blocking ATCase activity can slow tumor growth in cancer.

EXPLANATION: Discussions of DNA in earlier grades have been limited to discussions of genes and the role of genes in the appearance and activities of an organism. Biology 1 represents a student's introduction to a molecular model of a DNA as well as the organization of DNA into genes and genes subsequently into chromosomes. Students should address interactions between genes in proteins which regulate both the shape and reproduction of DNA molecules. It is important to note that not all segments of DNA are transcribed for proteins. Portions of DNA are involved in regulating the expression of genes or affect the structure of the chromosomes (replication origin, centromeres, and telomeres).
