Genome-wide Association Studies-GWAS

Manchmal können SNPs innerhalb eines Haplotyp-Blocks dabei helfen, ein Gen zu finden, das für bestimmte Krankheiten - wie Hämophilie - verantwortlich ist. Um solchen Genen auf die Spur zu kommen, können Forscher Populationsstudien durchführen.
Zum Beispiel werden genomweite Assoziationsstudien (GWAS) durchgeführt, um bestimmte genetische Variationen mit einer Krankheit in Verbindung zu bringen.
Bei diesem Ansatz verwenden die Forscher zwei große Studiengruppen: Menschen mit der interessierenden Krankheit und eine ähnliche Gruppe von Menschen ohne die Krankheit.
DNA-Proben von jedem Studienteilnehmer werden isoliert und sequenziert, bevor die Genome auf SNPs gescannt werden.
Wenn bestimmte SNPs bei Personen mit der Krankheit häufiger auftreten als bei gesunden Personen, dann spricht man von diesen SNPs als mit der Krankheit assoziiert.
Es ist zwar möglich, dass diese SNPs die Erkrankung verursachen, aber es ist wahrscheinlicher, dass sie Teil eines Haplotyp-Blocks sind, der die kausale Variante enthalten kann.

Genomweite Assoziationsstudien oder GWAS werden verwendet, um zu identifizieren, ob häufige SNPs (Single Nucleotide Polymorphisms) mit bestimmten Krankheiten in Verbindung stehen. Wenn spezifische SNPs häufiger bei Personen mit einer bestimmten Krankheit im Vergleich zu Personen ohne die Krankheit beobachtet werden, wird gesagt, dass diese SNPs mit der Krankheit assoziiert sind. Zur Überprüfung der Wahrscheinlichkeit, dass das Allel mit der Krankheit assoziiert ist, wird eine Chi-Quadrat-Analyse durchgeführt.

GWAS erfordert nicht die Identifizierung des Zielgens, das am Krankheitsphänotyp beteiligt ist. Es identifiziert den SNP, der mit der Krankheit assoziiert ist, und hilft dabei, die Individuen zu identifizieren, die ein Risiko tragen. Genome-wide association studies, or GWAS, look for common SNPs, or single nucleotide polymorphisms, that may be linked to disease. Researchers compare how often a SNP appears in people with a disease and in people without it. When a SNP is seen more often in the disease group, it is considered associated with that disease.

GWAS uses chi-square analysis to test the chance that an allele is linked to the disease. This approach does not require scientists to first find the target gene behind the disease trait. Instead, it helps point to the SNP tied to risk and can help identify individuals who may be at higher risk.

This method can make gene finding easier, since searching for the target gene can take a long time and may not be accurate in many cases. GWAS has been used to find SNPs linked to an increased risk of myocardial infarction. It has also identified SNPs associated with type 2 diabetes, Parkinson's disease, other heart disorders, obesity, Crohn's disease, and prostate cancer.
