Anaphase Promoting Complex

Der erfolgreiche Abschluss der Anaphase hängt auch von der Aktivität einer Ubiquitin-Ligase ab, die als Anaphase-fördernder Komplex oder Zyklosom APC/C bezeichnet wird.
Während des Übergangs in die Anaphase bindet das Protein Cdc20 an APC/C und aktiviert es. Das aktivierte APC/C ubiquitiniert die regulatorischen Proteine S-Cycline und M-Cycline und zielt so auf den proteasomalen Abbau ab.

Der schrittweise Abbau spezifischer Proteine ist notwendig für den Fortschritt und Abschluss des Zellzyklus. Solche Proteine werden durch Ubiquitin-Ligasen ubiquitiniert und anschließend durch das Proteasom zerstört. Der SCF (Skp1/Cullin/F-box) und der anaphase-fördernde Komplex (APC) sind zwei wichtige Ubiquitin-Ligasen, die an der Zellzyklusprogression beteiligt sind. Während SCF während des gesamten Zellzyklus aktiv ist, wird APC während des Übergangs von der Metaphase zur Anaphase aktiviert. Cdc20 oder Cdh1 binden an den APC und aktivieren ihn, was zur Bildung der APC^Cdc20- und APC^Cdh1-Komplexe führt. Cdc20 und Cdh1 helfen auch, dem APC seine Substrate zu identifizieren.

APC^Cdc20 spielt eine wesentliche Rolle beim Übergang von der Metaphase zur Anaphase. Die Schwesterchromatiden werden durch Kohäsine zusammengehalten. Die Kohäsinringe müssen durch das Enzym Separase gespalten werden, damit die Schwesterchromatiden während der Anaphase zu den entgegengesetzten Polen wandern können. Die Aktivität der Separase wird jedoch durch das Protein Securin gehemmt. APC^Cdc20 ubiquitiniert Securin, was zu dessen proteasomalem Abbau führt. Dies führt zur anschließenden Aktivierung der Separase. APC^Cdc20 ubiquitiniert auch mitotische Cycline, was wiederum die zyklin-abhängigen Kinasen oder Cdks inaktiviert. Die Inaktivierung der Cdks und die anschließende Dephosphorylierung der Cdk-Substrate sind notwendig für den Abschluss der Zellteilung.

The anaphase-promoting complex, or APC, helps control cell cycle progression by destroying specific proteins at the right time. These proteins are tagged with ubiquitin by ubiquitin ligases and then broken down by the proteasome. The SCF (Skp1/Cullin/F-box) complex and APC are two major ubiquitin ligases involved in this process.

SCF stays active throughout the cell cycle, but APC turns on during the metaphase-to-anaphase transition. APC becomes active when Cdc20 or Cdh1 binds to it. These factors form APC Cdc20 and APC Cdh1 complexes, and they also help APC recognize its target proteins.

APC Cdc20 is essential for sister chromatid separation. Sister chromatids are held together by cohesins, which form rings that must be cut so the chromatids can move to opposite poles during anaphase. Separase is the enzyme that breaks the cohesin rings, but securin keeps separase inactive until APC Cdc20 ubiquitinates securin and sends it for proteasomal degradation.

APC Cdc20 also ubiquitinates mitotic cyclins. This lowers cyclin-dependent kinase, or Cdk, activity. The loss of Cdk activity and the later dephosphorylation of Cdk substrates are needed to finish cell division.
