Incremental Stimulation Technique

Incremental Stimulation Technique is a behavioral method that gradually increases the intensity, complexity, or duration of a stimulus to encourage a targeted response while limiting abrupt changes that may disrupt behavior. The procedure presents manageable stimulus steps, observes the individual’s response, and advances only when the current level is tolerated or produces the desired behavior, often with reinforcement for successful performance. In behavioral research and training, this approach can support stimulus control, shape successive approximations, and improve participation in challenging tasks. It also helps investigators examine response thresholds, learning patterns, and the conditions under which behavior changes as stimulation increases.

This video demonstrates an in vitro technique to stimulate lymphocytes via non-typeable Haemophilus influenzae (NTHi) and to assess the immune response. Incubating isolated peripheral blood mononuclear cells (PBMCs) with NTHi causes the bacterial cells to stimulate T lymphocytes, leading to cytokine production. Upon inhibiting the secretion of cytokines, immunofluorescence staining is performed to detect the accumulated intracellular cytokines in stimulated T lymphocytes.
