Plasma Kynurenine Measurement

Plasma kynurenine measurement is the quantitative analysis of kynurenine, a major metabolite in the tryptophan degradation pathway, in blood plasma. In this method, plasma is separated from whole blood, kynurenine is isolated from other components through chromatographic separation, and its concentration is determined by comparison with calibrated analytical standards, often using liquid chromatography with optical or mass-spectrometric detection. In behavioral research, these measurements help assess links between tryptophan metabolism, immune signaling, and neuroactive metabolites that may influence mood, cognition, stress responses, and behavior. The approach supports biomarker studies and mechanistic investigations in both animal models and human research.

Here we utilize reducible, membrane impermeant biotinylation reagents to measure the endocytic rate of the dopamine transporter (DAT), a polytopic membrane protein. The method facilitates a straightforward approach to measuring the endocytic rate of most plasma membrane proteins.
