Estrogen Receptor Agonist Treatment

Estrogen receptor agonist treatment is the use of compounds that activate estrogen receptors to reproduce or enhance signaling normally triggered by endogenous estrogens, making it an important tool in biochemical research and medicine. After entering target cells, an agonist binds estrogen receptor proteins, promotes receptor activation and dimerization, and enables receptor complexes to interact with estrogen-responsive DNA elements and regulate gene transcription. These treatments help investigators examine hormone-dependent pathways, cellular differentiation, and gene regulation, while clinical applications can address conditions involving insufficient estrogen signaling. Experimental outcomes depend on receptor subtype, ligand selectivity, dose, and tissue context.

Estrogen Receptor-Reporter Activity Assay to Screen Phytoestrogen Estrogenic Activity

2025

This video describes a cell-based reporter assay to determine the estrogenic activity of plant-derived compounds. A compound with estrogen-like activity mimics its binding to estrogen receptors and drives downstream reporter enzyme gene transcription. The reporter enzyme generates bioluminescence in a reaction, indicating the estrogenic activity of the compound.

Quantifying Agonist Activity at G Protein-coupled Receptors

A method for estimating the affinity constant of an agonist for the active state (Kb) of a G protein-coupled receptor is described. The analysis provides absolute or relative measures of Kb depending on whether constitutive receptor activation is measurable. Our method applies to various responses downstream from receptor activation.

2014

Molecular signaling through both estrogen and microRNAs are critical in breast cancer development and growth. Estrogen activates the estrogen receptors, which are transcription factors. Many transcription factors can regulate the expression of microRNAs, and estrogen-regulated microRNAs can be profiled using different large-scale techniques.

Treatment for Pulmonary Arterial Hypertension: Prostacyclin Receptor Agonists

2024

Prostacyclin receptor agonists are a class of therapeutic agents integral to managing pulmonary arterial hypertension (PAH). These drugs operate by mimicking the action of prostaglandin I2, or PGI2, a naturally occurring compound in the body. These agonists bind to the IPR receptor situated on the plasma membrane of the pulmonary artery smooth muscle cells. This binding triggers a cascade of reactions known as the GS-AC-cAMP-PKA pathway. This pathway results in the relaxation of smooth muscle...
