Fluorescent Protein Screening

Fluorescent protein screening is a method for identifying fluorescent protein variants with desired optical or functional properties, making it valuable for studying protein behavior and cellular processes in biochemistry. In a typical workflow, candidate genes are expressed in cells or other biological systems, exposed to an excitation wavelength, and evaluated by their emitted light; fluorescence intensity, color, brightness, stability, or activity can guide selection of improved variants. Screening supports protein engineering and the development of genetically encoded reporters, biosensors, and labels for tracking molecular interactions, localization, and biochemical changes. These tools improve live-cell imaging and enable quantitative studies of biological mechanisms.

A procedure for liquid-based culturing and dispensing of C. elegans strains expressing fluorescent reporter proteins is described that does not require expensive sorting equipment. This approach can be applied to numerous inducible C. elegans genes for drug discovery or biosensing of contaminants.

The present protocol describes a procedure to perform fluorescent size exclusion chromatography (FSEC) on membrane proteins to assess their quality for downstream functional and structural analysis. Representative FSEC results collected for several G-protein coupled receptors (GPCRs) under detergent-solubilized and detergent-free conditions are presented.
