Glow Discharge Grid

A glow discharge grid is an electron microscopy support grid treated with a low-pressure plasma to modify its surface, an important step for preparing biological specimens. In the discharge, an applied electric field ionizes a gas, and energetic ions and electrons bombard the grid, removing contaminants and introducing polar surface groups that increase hydrophilicity. The resulting surface promotes more uniform spreading and adsorption of protein solutions, nucleic acids, and other biochemical samples. By improving specimen distribution and reducing uneven background, glow discharge treatment supports negative-stain electron microscopy and cryo-electron microscopy, contributing to more reliable imaging and structural analysis.

2011

Visualizing protein samples by negative stain electron microscopy (EM) has become a popular structural analysis method. It is useful for quantitative structural analysis, such as calculating a 3D reconstruction of the molecules being studied, and also for qualitative examination of the quality of protein preparations. In this article we present detailed protocols for preparing the EM grids, staining the sample and visualizing the sample in an electron microscope. Novice users can follow these...

RC Circuits: Discharging A Capacitor

2025

One of the applications of an RC circuit is the relaxation oscillator. The relaxation oscillator comprises a voltage source, a capacitor, a resistor, and a neon lamp. The lamp acts like an open circuit (infinite resistance) until the potential difference across the neon lamp reaches a specific voltage. At that voltage, the lamp acts like a short circuit (zero resistance), and the capacitor discharges through the neon lamp and produces light. Once the capacitor is fully discharged through the...
