Russian Wheat Aphid

Russian wheat aphid is a phloem-feeding insect pest, Diuraphis noxia, that damages wheat and other grasses by extracting nutrients and altering host physiology. Using piercing-sucking stylets, it penetrates vascular tissue and delivers saliva containing proteins and effectors that influence plant signaling, defense chemistry, and cellular metabolism, contributing to chlorosis, leaf rolling, and reduced growth. Biochemical study of these interactions examines changes in enzymes, antioxidants, metabolites, and stress-response pathways in both aphid and plant. This knowledge supports identification of resistance mechanisms, development of biochemical markers, and improved strategies for breeding and managing aphid-resistant cereal crops.

Aphids are effective transmitters of plant viruses. Aphid microinjection of virus, the procedure we will show you today, is a technique allowing researchers to inject virus directly into the hemocoel of the aphid, bypassing the gut, one of the 2 major barriers for virus transmission in a circulative manner. The same technique is also used to inject dsRNA for RNAi.

A protocol of whole-mount immunostaining on aphid embryos is presented, in which critical conditions for decreasing background staining and increasing tissue permeability are addressed. These conditions are specifically developed for effective detection of protein expression in the embryonic tissues of aphids.

Film Extrusion of Crambe abyssinica/Wheat Gluten Blends

The side stream from plant oil production of Crambe abyssinica has limited value. The purpose of this study was to find methods for extruding materials based on this side stream, demonstrating that products with a higher value can be produced. The extrudates were found to have promising properties.

Presented here is a protocol for the determination of the absorption, translocation, and distribution of imidacloprid in wheat under hydroponic conditions using liquid chromatography-tandem mass spectrometry (LC-MS-MS). The results showed that imidacloprid can be absorbed by wheat, and imidacloprid was detected in both the wheat roots and leaves.
