Grid Screening Automation

Grid screening automation is the use of robotic systems and software to test many biochemical conditions in a structured grid, improving the speed, consistency, and scale of experimental screening. The system typically dispenses samples and reagents into defined combinations, controls variables such as concentration, pH, or temperature, and records results through automated imaging or analytical measurements. In biochemistry, this approach helps identify promising conditions for protein crystallization, assay development, compound screening, and sample preparation. Automated data capture and comparison reduce manual error, support systematic optimization, and enable researchers to evaluate broad experimental spaces more efficiently.

This protocol shows how to set up a standard Leginon collection and Smart Leginon Autoscreen to automate this process. This protocol can be applied to the majority of cryoEM holey foil grids.

Development of automated imaging and analysis for zebrafish chemical screens.

We report the development of a system for automated imaging and analysis of zebrafish transgenic embryos in multiwell plates. This demonstrates the ability to measure dose dependent effects of a small molecule, BCI, on Fibroblast Growth Factor reporter gene expression and provide technology for establishing high-throughput zebrafish chemical screens.

Quantitative and Automated High-throughput Genome-wide RNAi Screens in C. elegans

We describe a protocol using C. elegans and RNAi feeding libraries that allows automated measurement of multiple parameters such as fluorescence, size and opacity of individual worms in a population. We give one example of a screen to identify genes involved in anti-fungal innate immunity in C. elegans.

Automated Modular High Throughput Exopolysaccharide Screening Platform Coupled with Highly Sensitive Carbohydrate Fingerprint Analysis

We present an automated modular high-throughput-method for the identification and characterization of microbial exopolysaccharides in small scale. This method combines a fast preselection to analyze the total amount of secreted polysaccharides with a detailed carbohydrate fingerprint to enable the fast screening of newly isolated bacterial strains or entire strain collections.
