High-ph Reversed Phase

High-pH reversed-phase chromatography is a liquid chromatography technique that separates molecules by hydrophobicity under alkaline mobile-phase conditions, making it valuable for biochemical analysis. Samples pass through a nonpolar stationary phase while an aqueous-organic solvent gradient elutes compounds according to their interactions with the stationary material; elevated pH changes analyte ionization and separation selectivity. The method is widely used to fractionate peptides and proteins, particularly before mass spectrometry-based proteomics, where it can reduce sample complexity and improve peptide identification. Its orthogonal selectivity compared with low-pH reversed-phase chromatography supports multidimensional separations and more comprehensive characterization of complex biological samples.
