Membrane Protein Purification

Membrane protein purification is the isolation of proteins embedded in lipid bilayers for biochemical, structural, and functional analysis. Because these proteins contain hydrophobic transmembrane regions, detergents or other membrane-mimetic systems disrupt lipid interactions and keep the proteins soluble while preserving their native conformation. A typical workflow combines membrane fractionation, solubilization, clarification, and chromatographic separation, often followed by reconstitution into liposomes or nanodiscs. Purified membrane proteins enable studies of transport, signaling, enzymatic activity, and drug binding, while supporting structural methods such as cryo-electron microscopy and X-ray crystallography.

Detergents are used to purify the integral proteins of the membrane. The hydrophobic portion of the detergent can replace membrane phospholipids while solubilizing the membrane proteins.
