Biological Fluid Analysis

Biological fluid analysis is the systematic examination of fluids such as blood, urine, saliva, and cerebrospinal fluid to measure their chemical, cellular, and molecular composition. Samples are collected, prepared, and analyzed using approaches such as microscopy, centrifugation, spectroscopy, immunoassays, or biosensors, which detect target molecules or cells through physical, chemical, or binding-based signals. In bioengineering, these measurements support biomarker discovery, disease monitoring, pharmacokinetic studies, and evaluation of engineered tissues or medical devices. Careful control of sampling, calibration, and matrix effects improves accuracy and helps translate fluid-based measurements into noninvasive diagnostics and personalized treatment strategies.

Characterizing Extracellular Vesicles from Biological Fluids

Here, we demonstrate the characterization method for extracellular vesicles (EVs) collected from biological fluids, such as tears and saliva, of human subjects. The scanner used in this method is capable of detecting the phenotype, size, and total particle count of EVs from 1 µL of the sample.

Ivacaftor and ivacaftor-lumacaftor combination are two new CF drugs. However, there is still a dearth of understanding on their PK/PD and pharmacology. We present an optimized HPLC-MS technique for the simultaneous analysis of ivacaftor and its major metabolites, and lumacaftor.

Sample Preparation for Endopeptidomic Analysis in Human Cerebrospinal Fluid

A method for mass spectrometric analysis of endogenous peptides in human cerebrospinal fluid (CSF) is presented. By employing molecular weight cut-off filtration, chromatographic pre-fractionation, mass spectrometric analysis and a subsequent combination of peptide identification strategies, it was possible to expand the known CSF peptidome nearly ten-fold compared to previous studies.

Cilia-generated fluid flow in Kupffer’s Vesicle (KV) controls left-right patterning of the zebrafish embryo. Here, we describe a technique to modulate gene function specifically in KV cells. In addition, we show how to deliver fluorescent beads into KV to visualize fluid flow.

Synovial Fluid Analysis to Identify Osteoarthritis

Synovial fluid analysis under transmitted, polarized, and compensated light microscopy is used to evaluate the inflammatory or non-inflammatory nature of a sample through simple steps. It is particularly useful in osteoarthritis to detect calcium crystals and identify a more severe subset of osteoarthritis.
