Constitutive Cre Expression

Constitutive Cre expression is the continuous production of Cre recombinase in a genetically defined cell population, enabling permanent control of DNA rearrangement and gene function. Cre recognizes loxP sites and catalyzes site-specific recombination, excising, inverting, or activating a DNA segment depending on the orientation and arrangement of those sites. In bioengineering, constitutive Cre systems generate stable genetic modifications throughout development or across targeted tissues, supporting conditional knockout and reporter models, lineage tracing, and engineered cell-line construction. Because recombination can persist after Cre expression ends, these systems provide a durable way to study gene function, cell fate, and long-term biological outcomes.

Here, we describe the methodology used for generating L. panamensis and L. donovani strains expressing the gene for eGFP as a stable integrated transgene using the pLEXSY system. Transfected parasites were cloned by limiting dilution, and clones with the highest fluorescence intensity in both species were selected for further use in drug screening assays.

In this video, a genetically modified mouse pup carrying the Ccm2 gene flanked by loxP sites and Cre recombinase fused to an estrogen receptor is used to induce cerebral cavernous malformations (CCMs). The administration of 4-hydroxytamoxifen activates Cre in endothelial cells, leading to the deletion of Ccm2 and the formation of CCMs in the brain.
