Limbal Stem Cells

Limbal stem cells are tissue-resident stem cells located at the corneal limbus, the boundary between the cornea and conjunctiva, where they maintain and repair the transparent corneal epithelium. Within their specialized niche, these cells self-renew and generate transient amplifying progenitors that proliferate, migrate across the corneal surface, and differentiate into mature epithelial cells. In bioengineering, researchers use cultivated limbal cells, biomaterial scaffolds, and engineered substrates to support cell expansion and restore damaged ocular surfaces. These approaches may improve treatments for limbal stem cell deficiency caused by injury or disease while advancing studies of stem cell niches, tissue regeneration, and ocular implants.

An example of adenoviral gene therapy in the human diabetic organ-cultured corneas is presented towards the normalization of delayed wound healing and markedly reduced epithelial stem cell marker expression in these corneas. It also describes the optimization of this process in stem cell-enriched limbal epithelial cultures.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
