Macrophage Cell Assay

A macrophage cell assay is an experimental system used to measure how macrophages respond to defined biological stimuli, materials, or therapeutic candidates, making innate immune behavior assessable in the laboratory. In a typical assay, cultured macrophages are exposed to a controlled condition and evaluated through readouts such as phagocytosis, cytokine secretion, viability, or surface markers that reflect activation and polarization. In bioengineering, these assays help determine how biomaterials, drug-delivery systems, implants, or engineered tissue environments may regulate inflammation and macrophage phenotype. Results support material optimization, biocompatibility testing, and the design of therapies intended to guide immune responses and improve integration with host tissue.

An Assay to Study the Regulation of Macrophage Phagocytosis by Mesenchymal Stem Cells

This video demonstrates an assay to study mesenchymal stem cell (MSC) regulation of macrophage phagocytosis in a co-culture. Upon co-culturing MSC with macrophages, non-opsonized zymosan particles conjugated to a pH-sensitive fluorophore are added to be phagocytosed by the macrophages. With the increase in phagocytosis, the fluorescence intensity rises, allowing the determination of MSC-mediated regulation of phagocytic activity.

In this video, we demonstrate the efferocytosis assay to study the clearance of apoptotic cells using confocal microscopy. Efferocytosis is a multi-step process where apoptotic cells are engulfed by phagocytes and degraded within vesicles called efferosomes. This protocol provides a method to visualize internalized and non-internalized portions of individual apoptotic cells and quantify efferocytosis.
