Murine Airway Model

A murine airway model is an experimental system that uses mice, mouse-derived airway tissues, or engineered airway constructs to study respiratory structure, function, and disease. Researchers alter genetic, cellular, biochemical, or mechanical conditions and assess how airway epithelium, immune cells, mucus production, barrier integrity, and tissue remodeling respond. In bioengineering, these models help evaluate biomaterials, inhaled therapeutics, tissue-engineered constructs, and regenerative strategies under controlled biological conditions. Because murine systems support both in vivo studies and complementary ex vivo or in vitro platforms, they enable mechanistic investigation of airway injury and inflammation while providing evidence for refining treatments and designing more predictive human-relevant models.

This paper describes different methods of growing Pseudomonas aeruginosa biofilms on cultured human airway epithelial cells. These protocols can be adapted to study different aspects of biofilm formation, including visualization of the biofilm, staining of the biofilm, measuring the colony forming units (CFU) of the biofilm, and studying biofilm cytotoxicity.

2023

The murine intrapulmonary tracheal transplantation (IPTT) model is valuable for studying obliterative airway disease (OAD) after lung transplantation. It offers insights into lung-specific immunological and angiogenic behavior in airway obliteration after allotransplantation with high reproducibility. Here, we describe the IPTT procedure and its expected results.
