Microwell Array Culture

Microwell array culture is a bioengineering technique that organizes cells in many small, spatially defined wells to support controlled cell growth and formation of three-dimensional assemblies. By confining a known number of cells within each microwell, the method promotes cell-cell contact and aggregation while reducing variation in spheroid size; well geometry, seeding density, and culture conditions can further shape outcomes. This scalable platform is used to produce cell spheroids and organoid-like structures for tissue engineering, disease modeling, drug screening, and studies of cell behavior, helping researchers standardize 3D culture and improve reproducibility.

The development of microbial communities depends on a combination of factors, including environmental architecture, member abundance, traits, and interactions. This protocol describes a synthetic, microfabricated environment for the simultaneous tracking of thousands of communities contained in femtoliter wells, where key factors such as niche size and confinement can be approximated.

This manuscript introduces a robust method of fabricating concave microwells without the need for complex high-cost facilities. Using magnetic force, steel beads, and a through-hole array, several hundred microwells were formed in a 3 cm x 3 cm polydimethylsiloxane (PDMS) substrate.

Multi-electrode Array Recordings of Neuronal Avalanches in Organotypic Cultures

A robust way to study neuronal avalanches, i.e. scale-invariant spatio-temporal activity bursts, indicative of critical state dynamics in cortex. Avalanches emerge spontaneously in developing superficial layers of cultured cortex which allows for long-term measurements of the activity with planar integrated multi-electrode arrays (MEA) under precisely controlled conditions.
