Mitochondrial Superoxide

Mitochondrial superoxide is a reactive oxygen species generated within mitochondria and an important indicator of cellular redox balance, energy metabolism, and oxidative stress. It forms when electrons escaping the respiratory chain, particularly at complexes I and III, reduce molecular oxygen to superoxide; mitochondrial superoxide dismutase then converts much of it to hydrogen peroxide, which can participate in signaling or cause molecular damage. In bioengineering, measuring mitochondrial superoxide helps evaluate engineered cells, biomaterials, and therapeutic strategies that alter metabolism or protect mitochondrial function. These analyses support the design of more resilient cell systems and the study of disease-related oxidative injury.

Confocal Imaging of Single Mitochondrial Superoxide Flashes in Intact Heart or In Vivo

Confocal scanning microscopy is applied for imaging single mitochondrial events in perfused heart or skeletal muscles in live animal. Real-time monitoring of single mitochondrial processes such as superoxide flashes and membrane potential fluctuations enables the evaluation of mitochondrial function in a physiologically relevant context and during pathological perturbations.

Mitochondria contain several flavin-dependent enzymes that can produce reactive oxygen species (ROS). Monitoring ROS release from individual sites in mitochondria is challenging due to unwanted side reactions. We present an easy, inexpensive method for direct assessment of native rates for ROS release using purified flavoenzymes and microplate fluorometry.

In this article we describe an adapted relatively easy method using the fluorescence dye diaminofluorescein-2 diacetate (DAF-2DA) and dihydroethidium (DHE) for en face simultaneous detection and visualization of intracellular nitric oxide (NO) and superoxide anion (O2.−) respectively, in freshly isolated intact aortas of an obesity mouse model.
