Plasma Serum Analysis

Plasma and serum analysis is the measurement of cell-free blood-fluid components to characterize proteins, metabolites, electrolytes, hormones, and other biomarkers relevant to health and engineered biological systems. Plasma is collected from anticoagulated blood and retains clotting factors, whereas serum is obtained after blood clots and lacks most of these factors. In practice, samples are separated by centrifugation, then analyzed using biochemical, immunological, or instrument-based assays that quantify target molecules and reveal changes in concentration or activity. In bioengineering, these measurements support biomaterial evaluation, cell and tissue culture monitoring, disease modeling, drug testing, and development of diagnostic or therapeutic platforms by linking molecular readouts to biological function.

Several methods have been used for analyzing plasma lipoproteins; however, ultracentrifugation is still one of the most popular and reliable methods. Here, we describe a method regarding how to isolate lipoproteins from plasma using sequential density ultracentrifugation and how to analyze the apolipoproteins for both diagnostic and research purposes.
