Fluorescent Derivatization

Fluorescent derivatization is a chemical labeling technique that attaches a fluorescent group to a target molecule, making otherwise difficult-to-detect analytes measurable through emitted light. Typically, a fluorophore-containing reagent reacts with a specific functional group, such as an amine or sulfhydryl, to form a stable covalent derivative; excitation at a selected wavelength then produces fluorescence at a longer wavelength. In biological techniques, this approach improves sensitivity and selectivity in chromatography, electrophoresis, and microscopy. It supports the identification and quantification of metabolites, peptides, proteins, and other biomolecules, particularly when native compounds have weak or no fluorescence.

Derivatization of Protein Crystals with I3C using Random Microseed Matrix Screening

This article presents a method to generate protein crystals derivatized with I3C (5-amino-2,4,6-triiodoisophthalic acid) using microseeding to generate new crystallization conditions in sparse matrix screens. The trays can be set up using liquid dispensing robots or by hand.

We describe a method protocol for the GC-based analysis of the aldonitrile acetate derivatives of glucosamine and muramic acid extracted from soil. For elucidation of the chemical mechanism, we also present a strategy to confirm the structure of the derivative and the ion fragments formed upon electron ionization.
