Fluorescent Protein Plasmids

Fluorescent protein plasmids are circular DNA vectors that encode fluorescent proteins, enabling researchers to visualize cellular structures, gene expression, and biological processes. After introduction into cells, the plasmid uses regulatory elements such as a promoter to drive fluorescent protein production; the protein then absorbs light at a specific excitation wavelength and emits light at a longer wavelength. In biological techniques, these plasmids serve as reporter systems for monitoring gene activity, tagging proteins, tracking cell populations, and assessing transfection or expression efficiency. Their compatibility with live-cell imaging makes them valuable for studying protein localization, cell dynamics, and disease-related mechanisms.

A method of observing individual DNA molecules in live cells is described. The technique is based on the binding of a fluorescently tagged lac repressor protein to binding sites engineered into the DNA of interest. This method can be adapted to follow many recombinant DNAs in live cells over time.
