Germline Transformation

Germline transformation is a biological technique that introduces genetic material into germ cells or early embryos so the resulting genetic change can be inherited by future generations. The process typically delivers a transgene into cells that contribute to the reproductive lineage, where it may integrate into the genome and become transmitted through gametes, distinguishing it from nonheritable somatic modification. In biological research, germline transformation generates transgenic organisms for studying gene function, regulation, development, and disease mechanisms. It also supports experimental breeding and the development of engineered model systems, making it valuable for investigating how specific genetic changes influence traits across generations.

We present an immunofluorescence imaging-based method for spatial and temporal localization of active ERK in the dissected C. elegans gonad. The protocol described here can be adapted for visualization of any signaling or structural protein in the C. elegans gonad, provided a suitable antibody reagent is available.
