Thermal Anesthesia Method

Thermal anesthesia is a biological technique that uses controlled temperature changes to produce temporary, reversible immobilization in experimental organisms, reducing the need for chemical anesthetics during handling. Cooling or warming alters metabolic activity and suppresses neural and muscular function, with the effect depending on the organism, exposure temperature, and treatment duration. Researchers apply this method when collecting, transferring, imaging, or performing procedures on small ectothermic animals and other temperature-sensitive specimens. Careful control is essential because excessive or prolonged exposure can cause physiological stress, injury, or death, whereas appropriate conditions allow recovery and support reliable experimental results.

Local and Global Methods of Assessing Thermal Nociception in Drosophila Larvae

In this article, we demonstrate assays to study thermal nociception in Drosophila larvae. One assay involves spatially-restricted (local) stimulation of thermal nociceptors1,2 while the second involves a wholesale (global) activation of most or all such neurons3. Together, these techniques allow visualization and quantification of the behavioral functions of Drosophila nociceptive sensory neurons.
