Two-dimensional Nmr Spectrum

A two-dimensional NMR spectrum is a map of nuclear magnetic resonance signals across two frequency dimensions, allowing chemically similar resonances to be separated and relationships between nuclei to be examined. It is generated with a pulse sequence that encodes magnetization during an indirect-evolution period, transfers coherence between coupled or spatially proximate nuclei, and uses Fourier transformation to produce diagonal peaks and cross-peaks. In biological research, spectra such as COSY, TOCSY, and NOESY support resonance assignment, protein and nucleic-acid structure determination, and analysis of molecular interactions and dynamics. These measurements help characterize biomolecules in solution and evaluate conformational or binding changes.

The 1D NMR spectrum of large and complex molecules like natural products has complicated splitting patterns and overlapping signals, which can be easily interpreted using 2-dimensional (2D) NMR. Unlike 1D NMR, 2D NMR has two frequency axes that provide the coupling information between the nucleus A and nucleus B in a molecule. The process from which 2D spectra are obtained has four steps. The first step is the preparation period, during which nucleus A is excited with a radiofrequency pulse.

Hyperpolarized Xenon for NMR and MRI Applications

The production of hyperpolarized xenon by means of spin exchange optical pumping (SEOP) is described. This method yields a ~10000-fold enhancement of the nuclear spin polarization of Xe-129 and has applications in nuclear magnetic resonance spectroscopy and imaging. Examples of gas phase and solution state experiments are given.
