Vlp Capture Assay

A VLP capture assay is an immunoassay used to detect and quantify virus-like particles, noninfectious structures that mimic the organization of viruses without containing a complete viral genome. In the assay, an immobilized capture antibody binds surface features of the VLP, while a labeled detection antibody recognizes another accessible epitope to generate a measurable signal related to particle abundance. This technique supports VLP characterization, production monitoring, quality assessment, and vaccine research by providing a practical measure of particle levels in biological or manufacturing samples. Results can help evaluate consistency and guide the development of VLP-based vaccines and delivery systems.

Here, we present a protocol to detect neutralization epitopes on antigen-displaying virus-like particles (VLPs). Immunoprecipitation of the human immunodeficiency virus (HIV)-derived VLPs is performed using envelope glycoproteins-specific monoclonal antibodies coupled to protein G-conjugated magnetic beads. Captured VLPs are subsequently subjected to SDS-PAGE and Western blot-analysis employing viral core protein Gag-specific antibodies.

Conversion of a Capture ELISA to a Luminex xMAP Assay using a Multiplex Antibody Screening Method

An ELISA can be easily converted to a Luminex xMAP assay and, through the benefits of multiplexing, several antibodies can be screened simultaneously to identify an optimum antibody pair, resulting in increased sensitivity and dynamic range, while reducing assay cost.
