Electrophoresis Voltage Settings

Electrophoresis voltage settings are the electrical conditions used to drive charged molecules through a gel or other separation medium, directly influencing separation speed, resolution, and sample integrity. An applied voltage creates an electric field that moves molecules according to their charge, size, and interactions with the matrix; higher voltages generally increase migration rate but can generate heat, distort bands, or reduce resolution. In biological techniques, selecting an appropriate voltage supports reliable analysis of DNA, RNA, and proteins by balancing run time with clear band separation. Careful optimization improves reproducibility and helps prevent overheating, diffusion, and incomplete separation.

Source: Laboratory of Dr. B. Jill Venton - University of Virginia Capillary electrophoresis (CE) is a separation technique that separates molecules in an electric field according to size and charge. CE is performed in a small glass tube called a capillary that is filled with an electrolyte solution. Analytes are separated due to differences in electrophoretic mobility, which varies with charge, solvent viscosity, and size. Traditional electrophoresis in gels is limited in the amount of voltage...

Two-Dimensional Gel Electrophoresis

Two-dimensional gel electrophoresis (2DGE) is a technique that can resolve thousands of biomolecules from a mixture. This technique involves two distinct separation methods that have been coupled together: isoelectric focusing (IEF) and sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). This physically separates compounds across two axes of a gel by their isoelectric points (an electrochemical property) and their molecular weights. The procedure in this video covers the main...

DNA Gel Electrophoresis

DNA gel electrophoresis is a technique used for the detection and separation of DNA molecules. An electric field is applied to a gel matrix comprised of agarose, and within the gel, charge particles will migrate and separate based on size. The negatively charged phosphates of the DNA backbone cause DNA fragments to move toward the anode - a positively charged electrode. The video explains the mechanism by which DNA fragments are resolved on an agarose gel, and it provides a step-by-step...

Native polyacrylamide gel electrophoresis is a fundamental tool for analyzing RNA-protein interactions. Traditionally most experiments have used vertical gels. However, horizontal gels provide several advantages, such as the opportunity to monitor complexes during electrophoresis. We provide a detailed protocol for generating and using horizontal native gel electrophoresis.

The movement of electrons in a conductor requires some form of energy or work, usually provided by an external force, like a battery. This force is called the electromotive force or voltage. The voltage between two points, referred to as points "a" and "b," in an electric circuit is the energy (or work) needed to move a unit charge from point "a" to point "b," and this relationship is expressed mathematically as In this equation, "w" represents the energy measured in joules (J), and "q"...
