5-aminolevulinic Acid

5-aminolevulinic acid (5-ALA) is a metabolic precursor in the biosynthesis of heme, the iron-containing component of hemoglobin and many cellular enzymes. In mitochondria, 5-ALA synthase forms 5-ALA from glycine and succinyl-CoA, after which enzymatic reactions convert it through porphyrin intermediates into protoporphyrin IX and, ultimately, heme. Administered 5-ALA can increase protoporphyrin IX accumulation in certain tissues, enabling fluorescence-based detection of abnormal cells and photodynamic therapy when activated by light. Its study therefore connects cellular metabolism, mitochondrial function, porphyrin biochemistry, and biomedical applications such as tumor visualization and treatment.

This manuscript describes a protocol to study the antimicrobial effect of 5-aminolevulinic acid-mediated photodynamic therapy (ALA-PDT) on a Staphylococcus aureus biofilm. This protocol can be used to develop an in vitro model to study the treatment of bacterial biofilms with PDT in the future.
