Root Hair Analysis

Root hair analysis is the study and measurement of the fine, tubular extensions that develop from root epidermal cells, revealing how plants acquire resources and interact with their surroundings. Using microscopy and image-based measurements, researchers assess root hair density, length, distribution, and branching, often under controlled nutrient, water, or environmental conditions. These traits increase the root surface area available for absorbing water and mineral nutrients and can reflect plant responses to soil conditions. In biology, root hair analysis supports research on plant development, nutrient uptake, stress adaptation, and beneficial interactions with soil microorganisms, helping clarify how root systems function at cellular and ecological scales.

Imaging the Root Hair Morphology of Arabidopsis Seedlings in a Two-layer Microfluidic Platform

This article demonstrates how to culture Arabidopsis thaliana seedlings in a two-layer microfluidic platform that confines the main root and root hairs to a single optical plane. This platform can be used for real-time optical imaging of fine root morphology as well as for high-resolution imaging by other means.

Cortisol (CORT) accumulates in the growing hair shaft of humans and nonhuman primates. We describe methods for extracting and analyzing hair CORT with high precision and sensitivity. Measurement of hair CORT is particularly well-suited for assessing chronic stress over periods of weeks to months.

Hair cortisol concentration analysis provides an alternative to traditional measures of cortisol; however, to collect hair samples from African Americans, scientists need to be culturally informed and competent. The purpose of this protocol is to demonstrate a culturally informed technique to collect hair samples for cortisol analysis from African Americans.

Current methods of analyzing patients’ adherence to complex drug resistant-tuberculosis (DR-TB) regimens can be inaccurate and resource-intensive. Our method analyzes hair, an easily collected and stored matrix, for concentrations of 11 DR-TB medications. Using LC-MS/MS, we can determine sub-nanogram drug levels that can be utilized to better understand drug adherence.
