Alkaline Treatment

Alkaline treatment is a chemical method that uses a basic solution to modify biological materials, making selected components easier to separate, analyze, or use in subsequent processes. High pH promotes reactions such as hydrolysis and saponification, which can break ester linkages, remove structural components, and disrupt interactions within complex biological matter; the result depends on the alkali, concentration, temperature, and exposure time. In biology, researchers use alkaline treatment to process biomass, improve access to cellular or structural constituents, and support downstream isolation and characterization. Careful control is important because excessive alkalinity can damage target molecules or alter their properties.

The alkaline comet assay measures DNA strand breaks in eukaryotic cells. By adding an Endonuclease III or human 8-oxoguanine-DNA-N-glycosylase digestion step, the assay can efficiently detect oxidative DNA damage. We describe methods for using these assays to detect DNA damage in rat liver.
