Anti-cd3 Stimulation

Anti-CD3 stimulation is a laboratory method for activating T lymphocytes by targeting the CD3 complex associated with the T-cell receptor, making it useful for studying immune-cell function. Typically, anti-CD3 antibodies bind CD3 on the T-cell surface and, when immobilized or combined with crosslinking and co-stimulatory signals, initiate receptor-associated signaling that promotes T-cell activation, proliferation, and cytokine production. Researchers use this approach to analyze T-cell responses, compare immune-cell phenotypes, evaluate immunomodulatory treatments, and generate activated T cells for further experiments. Its controlled activation provides a reproducible model for investigating antigen-receptor signaling and cellular immunity.

Generating Anti-Angiogenic Tumor-Associated Neutrophils

2025

This video demonstrates an assay to generate anti-angiogenic tumor-associated neutrophils (TANs). Upon taking a murine melanoma tumor containing pro-angiogenic TANs, the single cells are isolated using fluorescence-activated cell sorting (FACS) and treated with an inhibitor of nicotinamide phosphoribosyl transferase (NAMPT) to reduce the capacity to stimulate angiogenesis.

Neutrophils play an important role not only in host defense against invading microorganisms, but are also involved in the immune surveillance of tumor cells. Here, we describe techniques related to the isolation of neutrophils with anti-tumor properties and methods for monitoring anti-tumor neutrophil function in vitro and in vivo.
