Antigen Specific T Cells

Antigen-specific T cells are lymphocytes that recognize particular molecular targets, enabling the adaptive immune system to respond selectively to infections, tumors, and other biological challenges. Their T-cell receptors bind antigen-derived peptides displayed by major histocompatibility complex molecules on antigen-presenting cells; with appropriate co-stimulatory signals, this interaction triggers clonal expansion, cytokine production, and differentiation into effector or memory cells. Studying these cells helps explain immune specificity and long-term protection, while methods that identify, isolate, or measure them support vaccine development, infection monitoring, cancer immunotherapy, and investigation of autoimmune disease.

Fluorescently-labeled viable cells are selected using flow cytometry, and the cells in the different cell cycle stages are identified by their DNA content using antibodies against cell proliferation-associated nuclear protein and a nucleic acid stain.

This video describes a protocol for isolating rare-antigen-specific CD8+ T cells using magnetic nanoparticle-based artificial antigen-presenting cells (aAPCs) functionalized with antigenic peptide-loaded MHC-Igs and anti-CD28 antibodies.
