Warm Media Recovery

Warm Media Recovery is a biological technique that helps stressed or genetically manipulated cells regain viability by placing them in prewarmed growth medium under controlled conditions. In bacterial transformation, cells are transferred to warm, nutrient-rich medium after DNA uptake, allowing membrane repair, metabolic recovery, and expression of newly acquired antibiotic-resistance genes before selective plating. This step improves transformation efficiency and increases the number of colonies available for analysis. Warm Media Recovery is therefore important in molecular biology workflows involving plasmid introduction, genetic engineering, and selection of successfully transformed microorganisms.

The goal of this procedure is to easily and rapidly produce a microfluidic device with customizable geometry and resistance to swelling by organic fluids for oil recovery studies. A polydimethylsiloxane mold is first generated, and then used to cast the epoxy-based device. A representative displacement study is reported.

2013

The gradient echo memory is a protocol for storing optical quantum states of light in atomic ensembles. Quantum memory is a key element of a quantum repeater, which can extend the range of quantum key distribution. We outline the operation of the scheme when implemented in a 3-level atomic ensemble.

(2017)This video demonstrates the recovery of infectious Zika virus using electroporation of in vitro transcribed RNA. The process shows how synthetic RNA initiates infection and leads to viral replication in cultured cells.

Here we demonstrate the newly developed I•Cryo kit for mouse sperm cryopreservation. Two-cell stage embryo development with frozen-thawed sperm was improved consistently in 5 mouse strains with the use of this kit. Over a 1.5 year period, 49 genetically modified mouse lines were archived by sperm cryopreservation with the I•Cryo kit and later successfully recovered by IVF.

Reverse Genetics Mediated Recovery of Infectious Murine Norovirus

Noroviruses are a major cause of gastroenteritis yet molecular techniques for their characterisation are still relatively new. Here we report two different reverse genetics approaches for the efficient recovery of murine norovirus (MNV), the only member of this genus which can be propagated in cell culture.
