Caspase 3 Cleavage

Caspase 3 cleavage is a proteolytic process that activates the executioner caspase-3 enzyme during apoptosis, the regulated form of cell death. Initiator caspases cleave inactive procaspase-3 into active subunits, which then recognize protein substrates containing specific aspartate residues and dismantle essential cellular components. This activity contributes to hallmark apoptotic changes, including DNA fragmentation, cytoskeletal breakdown, and cellular shrinkage. Measuring cleaved caspase-3 with immunoblotting, immunostaining, or activity assays helps researchers identify apoptotic cells and evaluate cell death pathways in development, disease research, toxicology, and studies of anticancer treatments.

Assessing Caspase-Mediated Cleavage of Viral and Host Proteins in Virus-Infected Cells

2025

The video demonstrates a method to assess caspase-mediated protein cleavage in virus-infected cells. Post-viral infection, caspases get activated and degrade various host cellular proteins and viral nucleoproteins while specific caspase inhibitors prevent this degradation, as confirmed by western blotting.

Caspase-3/7 Assay: A Luminescence-Based Assay to Quantify Apoptosis by Measuring Caspase-3/7 Activities in Frog Skin Explants

2025

This video describes an in vitro Caspase-3/7 luminescent assay to quantify cell death or apoptosis. The assay measures luminescence produced following caspase-3/7 cleavage of a pro-luminescent DEVD substrate. The luminescence generated is proportional to caspase-3/7 activity.

2016

Myocardial infarction (MI) is not only followed by impaired cardiac function but also by apoptosis in the amygdala, a brain region involved in the behavioral consequences of MI. This protocol describes how to induce MI, collect amygdala tissue and measure caspase-3 activity, a marker of apoptosis, therein.

2013

Certain genetic perturbations or exposure to toxins can disrupt normal developmental processes leading to death of specific cell types. The analysis of activated Caspase 3 by whole-mount immunofluorescence in zebrafish embryos reveals stage- and tissue-specific localization of cells specifically undergoing apoptosis.

Assessment of the Activation of Multiple Caspases in Macrophages

2025

This video demonstrates the assessment of multiple caspase activations in bone marrow-derived macrophages through stimulation with the influenza virus. Following macrophage lysis, heat-inactivation of lysate, and centrifugation, intracellular caspases are collected for subsequent immunological analysis.
