Chromosome Spreading

Chromosome spreading is a cytogenetic technique that separates condensed chromosomes on a microscope slide so their number, size, and structure can be examined. Cells are typically arrested during metaphase, when chromosomes are highly condensed, then treated with a hypotonic solution to swell them before fixation and gentle dispersion onto the slide. The resulting chromosome preparation supports karyotyping and the detection of numerical or structural abnormalities, including changes in chromosome count, rearrangements, and large deletions. By making individual chromosomes accessible for microscopic or molecular analysis, chromosome spreading contributes to research in cell biology, genetics, developmental disorders, cancer, and reproductive biology.

Surface Spreading and Immunostaining of Yeast Chromosomes

A method for surface-spreading chromosomes from budding yeast is presented. This method is derived from a method previously described by Loidl and Klein. In addition, we demonstrate a procedure for immunostaining of spread chromosomes.

Mammalian meiosis can be examined by utilizing a technique to prepare meiotic chromosome spreads. Here, we demonstrate a method of preparing surface-spread nuclei from mouse spermatocytes.
