Clonogenic Survival Assay

The clonogenic survival assay is a cell-based method that measures the ability of a single cell to survive treatment and produce a viable colony, providing a long-term readout of reproductive survival. In this assay, cells are exposed to a defined condition, plated at low density, and cultured until surviving cells undergo repeated division to form colonies, which are then counted to calculate survival fractions. In biology, the method is used to assess cellular responses to radiation, chemotherapy, genetic manipulation, and other stresses. Its results help researchers compare treatment potency, identify changes in proliferative capacity, and investigate mechanisms of cell death and recovery.

2023

This video introduces a short-term survival assay that tests a treatment for acute toxicity or tests a genetic strain for resistance to a toxic condition. The example protocol shows an example survival assay in which worms are tested for their ability to resist oxidative stress induced by paraquat (PQ).
