Colony Pcr Verification

Colony PCR verification is a rapid molecular biology method used to determine whether individual bacterial colonies contain a DNA construct of the expected size and sequence. In this approach, a small amount of colony material serves as the template for polymerase chain reaction, where repeated cycles of denaturation, primer annealing, and extension amplify a selected region; the products are then separated by agarose gel electrophoresis and assessed by band size. Researchers use colony PCR to screen transformed cells after cloning, identify colonies carrying an insert, and reduce the number of samples requiring plasmid purification and sequencing. This efficient verification step supports genetic engineering, recombinant DNA studies, and routine strain construction.
