Constitutive Receptor Activity

Constitutive receptor activity is the ability of a cell-surface or intracellular receptor to adopt an active signaling state and trigger cellular responses without binding its usual ligand. This activity arises from a conformational equilibrium in which receptors spontaneously shift between inactive and active forms, with mutations, receptor abundance, or cellular conditions altering that balance. Constitutive signaling is particularly important in biology and pharmacology because it can influence gene expression, metabolism, proliferation, and other physiological processes. Studying it helps researchers identify activating mutations, distinguish agonists from inverse agonists, and understand how abnormal receptor activity contributes to disease.

2011

A method for estimating the affinity constant of an agonist for the active state (Kb) of a G protein-coupled receptor is described. The analysis provides absolute or relative measures of Kb depending on whether constitutive receptor activation is measurable. Our method applies to various responses downstream from receptor activation.

High-throughput Analysis of Mammalian Olfactory Receptors: Measurement of Receptor Activation via Luciferase Activity

2014

Olfactory receptor activation patterns encode odor identity, but the lack of published data identifying odorant ligands for mammalian olfactory receptors hinders the development of a comprehensive model of odor coding. This protocol describes a method to facilitate high-throughput identification of olfactory receptor ligands and quantification of receptor activation.

2025

This video describes a cell-based reporter assay to determine the estrogenic activity of plant-derived compounds. A compound with estrogen-like activity mimics its binding to estrogen receptors and drives downstream reporter enzyme gene transcription. The reporter enzyme generates bioluminescence in a reaction, indicating the estrogenic activity of the compound.
