Cystatin B Variants

Cystatin B variants are inherited changes in the CSTB gene that can alter the structure, production, or function of cystatin B, a protein that regulates cysteine proteases involved in intracellular protein turnover. Depending on their molecular effect, variants may reduce cystatin B stability, abundance, cellular localization, or inhibition of proteases such as cathepsins, disrupting proteolytic balance in cells. Studying these variants helps explain the biology of Unverricht–Lundborg disease and other conditions associated with CSTB dysfunction. Variant analysis also supports genetic diagnosis, pathogenicity assessment, genotype–phenotype research, and investigation of how protease regulation contributes to neuronal health and disease.

Detection of Rare Genomic Variants from Pooled Sequencing Using SPLINTER

Pooled DNA sequencing is a fast and cost-effective strategy to detect rare variants associated with complex phenotypes in large cohorts. Here we describe the computational analysis of pooled, next-generation sequencing of 32 cancer-related genes using the SPLINTER software package. This method is scalable, and applicable to any phenotype of interest.

Generation of Influenza Virus Escape Variants against Neutralizing Antibodies

This video demonstrates a technique for generating escape variants of neutralizing influenza virus monoclonal antibodies. Antibody-neutralized virus suspension is injected into embryonated chicken eggs to propagate escape variants selectively. This results in a virus population predominantly of escape variants, confirmed via a Hemagglutination assay.

We describe a method by which we identify critical residues required for the binding of human or murine monoclonal antibodies that target the viral hemagglutinin of influenza A viruses. The protocol can be adapted to other virus surface glycoproteins and their corresponding neutralizing antibodies.

Merging Absolute and Relative Quantitative PCR Data to Quantify STAT3 Splice Variant Transcripts

Tandem splicing events occur at sites less than 12 nucleotides apart. Quantifying ratios of such splice variants is feasible using an absolute quantitative PCR approach. This manuscript describes how splice variants of the gene STAT3, in which two splicing events results in Serine-701 inclusion/exclusion and α/β C-termini, can be quantified.
