Denaturation Annealing Extension

Denaturation, annealing, and extension are the three temperature-controlled stages of a polymerase chain reaction (PCR), the repeated cycle that selectively amplifies a target DNA sequence. During denaturation, heat separates double-stranded DNA into single strands; during annealing, cooling allows short primers to bind complementary target regions; and during extension, a heat-stable DNA polymerase adds nucleotides from each primer to synthesize new strands. Repeating these steps approximately doubles the target region during each cycle under suitable conditions. Together, the stages enable sensitive detection and analysis of genetic material in cloning, diagnostics, sequencing preparation, and molecular biology research.
