Dendritic Cell Infection

Dendritic cell infection is the entry and, for some pathogens, replication of microorganisms within dendritic cells, specialized antigen-presenting cells that initiate and regulate adaptive immune responses. Infection begins when pathogen-associated molecules interact with cell-surface receptors, triggering uptake into endosomes or the cytoplasm, where innate signaling and antigen processing can occur. Dendritic cells then display pathogen-derived peptides on major histocompatibility complex molecules and migrate to lymphoid tissues, although certain pathogens can disrupt maturation, antigen presentation, or antiviral defenses. Studying these interactions clarifies host-pathogen biology and supports vaccine development, infectious disease research, and strategies to improve immune-based therapies.

T Cells Capture Bacteria by Transinfection from Dendritic Cells

Here a protocol is presented to measure bacterial capture by CD4+ T cells which occurs during antigen presentation via transinfection from pre-infected dendritic cells (DC). We show how to perform the necessary steps: isolation of primary cells, infection of DC, DC/T cell conjugate formation, and measurement of bacterial T cell transfection.

Unlike cell-free HIV-1 particles, infected CD4+ T cells are effectively sensed by plasmacytoid dendritic cells (pDCs). This manuscript describes a method where peripheral blood mononuclear cells (PBMCs) or isolated pDCs are co-cultured with HIV-1 infected T cells to evaluate innate sensing by pDCs as assessed by release of type-I IFN.

siRNA-Based Inhibition of Autophagy in Herpes Simplex Virus-Infected Immature Dendritic Cells

This video demonstrates a small interfering RNA (siRNA)-based technique to interfere with autophagic flux. A siRNA targeting FIP200 mRNA is introduced into monocyte-derived immature dendritic cells (iDCs) using electroporation, causing the knockdown of FIP200 expression and leading to the inhibition of nuclear lamin disruption via autophagy. Upon adding herpes simplex virus (HSV), the nuclear egress of viral progeny is inhibited due to the absence of lamin disruption via autophagic flux.
