Dna Template Preparation

DNA template preparation is the process of obtaining and preparing a DNA molecule so it can serve as a reliable starting template for downstream molecular biology experiments. It typically involves cell lysis, DNA isolation, purification to remove proteins and reaction-inhibiting contaminants, and assessment of concentration and integrity; reverse transcription may be included when RNA is converted into complementary DNA. A clean, appropriately diluted template allows sequence-specific primers or other enzymes to copy, amplify, or read the target sequence efficiently. These steps support PCR, quantitative PCR, sequencing, cloning, genotyping, and diagnostic research, where template quality strongly affects accuracy and reproducibility.

We describe the process of isolating high purity herpesvirus nucleocapsid DNA from infected cells. The final DNA captured from solution is of high concentration and purity, making it ideally suited for high-throughput sequencing, high fidelity PCR reactions, and transfections to produce new viral recombinants.

Preparation and Use of Photocatalytically Active Segmented Ag|ZnO and Coaxial TiO2-Ag Nanowires Made by Templated Electrodeposition

Procedures are outlined to prepare segmented and coaxial nanowires via templated electrodeposition in nanopores. As examples, segmented nanowires consisting of Ag and ZnO segments, and coaxial nanowires consisting of a TiO2 shell and a Ag core were made. The nanowires were used in photocatalytic hydrogen formation experiments.

The combination of chromatin immunoprecipitation and ultra-high-throughput sequencing (ChIP-seq) can identify and map protein-DNA interactions in a given tissue or cell line. Outlined is how to generate a high quality ChIP template for subsequent sequencing, using experience with the transcription factor TCF7L2 as an example.
