Expansin Activity Assay

An expansin activity assay is a laboratory test that measures the ability of expansin proteins to loosen plant cell walls, helping researchers understand how these proteins influence cell growth and tissue development. The assay typically monitors the extension of isolated cell walls after expansin addition, often under acidic conditions, where expansins disrupt noncovalent interactions between cellulose and surrounding matrix polysaccharides without cleaving the polymers. Results can reveal protein activity, pH dependence, and differences among expansin isoforms. In biology, these measurements support studies of cell expansion, seed germination, fruit ripening, plant development, and the potential use of expansins in biomass processing.

The present report describes an in vitro enzymatic assay to measure phosphatidylethanolamine methyltransferase activity using Leishmania cell extracts. This assay is based on the transfer of a radioactive methyl group from S-[Methyl-3H]adenosyl-L-methionine onto endogenous phosphatidylethanolamine.
