Golgi Apparatus Mitosis

Golgi apparatus mitosis refers to the coordinated disassembly, partitioning, and reassembly of the Golgi apparatus during cell division, a process that helps daughter cells inherit this essential organelle. As mitosis begins, mitotic kinases such as Cdk1 trigger phosphorylation of Golgi structural and trafficking proteins, causing stacked cisternae to fragment into vesicles and membrane clusters. These Golgi-derived components are distributed between daughter cells and reorganize after cytokinesis as phosphorylation decreases and membrane tethering and fusion resume. Studying Golgi apparatus mitosis clarifies organelle inheritance, cell-cycle regulation, and how disruptions in membrane trafficking may contribute to developmental abnormalities and disease.

Properly folded and assembled proteins are selectively packaged into vesicles that exit the ER. Motor proteins transport these vesicles to the Golgi apparatus for adding modifications that make these proteins functional at their destination. The Golgi apparatus is a eukaryotic organelle that has a distinctive ribbon-like appearance. It is a primary sorting and dispatch station for cargo arriving from the ER. Newly arriving vesicles enter the cis face of the Golgi, closest to the ER, and are...

As they leave the Endoplasmic Reticulum (ER), properly folded and assembled proteins are selectively packaged into vesicles. These vesicles are transported by microtubule-based motor proteins and fuse together to form vesicular tubular clusters, subsequently arriving at the Golgi apparatus, a eukaryotic endomembrane organelle that often has a distinctive ribbon-like appearance. The Golgi apparatus is a major sorting and dispatch station for the products of the ER. Newly arriving vesicles enter...

The precise localization of Golgi residents is essential for understanding the cellular functions of the Golgi. However, conventional optical microscopy is unable to resolve the sub-Golgi structure. Here we describe the protocol for a conventional microscopy based super-resolution method to quantitatively determine the sub-Golgi localization of a protein.

Live Cell Imaging of Mitosis

Mitosis is a form of cell division in which a cell’s genetic material is divided equally between two daughter cells. Mitosis can be broken down into six phases, during each of which the cell’s components, such as its chromosomes, show visually distinct characteristics. Advances in fluorescence live cell imaging have allowed scientists to study this process in great detail, providing important insights into the biological control of this process and how it might go wrong in diseases such as...
