Human Skin Isolation

Human skin isolation is the laboratory process of separating skin tissue or its component cells from a tissue sample for study, culture, or downstream analysis. The procedure typically involves careful tissue dissection, removal of unwanted structures, and mechanical or enzymatic dissociation to release populations such as keratinocytes, fibroblasts, or immune cells, which can then be separated and maintained under controlled culture conditions. In biology, isolated skin cells support research on wound healing, tissue regeneration, barrier function, infection, aging, and skin disease. Reliable isolation preserves cell viability and identity, enabling experiments with primary cultures, engineered tissue models, and therapeutic development.

This manuscript describes a FACS-based protocol for isolation of papillary and reticular fibroblasts from human skin. It circumvents in vitro culture which was inevitable with the commonly used isolation protocol via explant cultures. The emanating fibroblast subsets are functionally distinct and display differential gene expression and localization within the dermis.
