Intracellular Calcium Dynamics

Intracellular calcium dynamics describes the changing concentration, distribution, and timing of calcium ions within cells, a process that coordinates many essential biological functions. Cells generate these signals by opening calcium-permeable channels in the plasma membrane or intracellular stores such as the endoplasmic reticulum, while pumps, exchangers, and buffering proteins restore low resting concentrations and shape local calcium patterns. The resulting calcium spikes, waves, and oscillations can regulate muscle contraction, secretion, gene expression, metabolism, and cell survival. In biology research, measuring these dynamics helps clarify how cells communicate and respond to stimuli and reveals disruptions associated with disease and dysfunction.

We demonstrate how changes in the intracellular free calcium concentration and synaptic efficacy can be simultaneously monitored in a ganglion preparation of Aplysia. We image intracellular calcium using a fluorescent dye, Calcium Orange, and induce and monitor synaptic transmission with sharp (intracellular) electrodes.

Source: Doser, R., et al. Subcellular Imaging of Neuronal Calcium Handling In Vivo. J. Vis. Exp. (2023)This video demonstrates in vivo imaging of subcellular calcium flux in the ventral nerve cord neurons of transgenic Caenorhabditis elegans strains. Two distinct calcium indicators are employed: a cytoplasmic indicator in one strain and a mitochondrial indicator in another. Neurons are observed under a fluorescence microscope to monitor real-time changes in fluorescence intensity within the...

Intracellular calcium recycling plays a critical role in regulation of systolic and diastolic function in cardiomyocytes. Here, we describe a protocol to evaluate sarcoplasmic reticulum Ca2+ reserve and diastolic calcium removal function in cardiomyocytes by a calcium imaging system.

Simultaneous Measurement of Intracellular Calcium and Membrane Potential in a Mouse Cerebral Endothelium

2025

The video demonstrates the simultaneous measurement of intracellular calcium levels and changes in membrane potential in endothelial cells of mouse cerebral artery segments. The isolated endothelial tube is loaded with a fluorescent calcium indicator and treated with a drug that induces intracellular calcium release, resulting in a change in membrane potential. The indicator's fluorescence enables the measurement of calcium levels, and a recording electrode is inserted into a cell to measure...
