Ion Flux Calibration

Ion flux calibration is the process of relating an instrument’s signal to the movement of ions across a membrane or through a biological sample, ensuring that measurements are accurate and comparable. Calibration typically uses reference solutions or standards with known ion concentrations or fluxes to establish response, correct baseline signals, and assess sensitivity under defined experimental conditions. In biology, calibrated ion-flux measurements support studies of membrane transport, electrophysiology, cellular signaling, and ion-channel activity. Reliable calibration improves interpretation of changes in intracellular or extracellular ions and strengthens comparisons across experiments, instruments, and biological systems.

We describe the ion-selective self-referencing microelectrode that allows the measurement of specific ion fluxes at single cells and tissues in vivo.

This protocol describes the use of the Ramsay assay to measure fluid secretion rates from isolated Malpighian (renal) tubules from Drosophila melanogaster. In addition, the use of ion-specific electrodes to measure sodium and potassium concentrations in the secreted fluid, allowing calculation of transepithelial ion flux, is described.
