Lineage Marker Injection

Lineage marker injection is a cell-labeling technique used to determine how individual cells and their descendants contribute to tissues during development, regeneration, or disease. Researchers introduce a visible tracer, fluorescent dye, or genetic label into selected cells, then follow the marker through cell divisions and tissue formation using imaging or molecular analysis. The resulting lineage map reveals patterns of cell fate, migration, proliferation, and differentiation. In biology, this approach helps test developmental models, identify progenitor populations, and clarify how tissues are organized and repaired, providing evidence that links early cellular behavior to later anatomical and functional outcomes.

Co-localization of Cell Lineage Markers and the Tomato Signal

We developed two sets of tracing combinations in Rosa26tdtomato (ubiquitously expressed in all cells)/Cre (specifically expressed in chondrocytes) mice: one with 2.3Col1a1-GFP (specific to osteoblasts) and one with immunofluorescence (specific to bone cells). The data demonstrate the direct transformation of chondrocytes into bone cells.

Traditional methods of assessing adipogenic differentiation are cheap and easy to use, but are not specific to changes in gene expression. We have developed an assay to quantify mesenchymal cell differentiation into mature adipocytes using a lineage specific marker. This assay has diverse applications across basic research and clinical medicine.

This protocol delineates a way to label and trace the fate of small groups of cells zebrafish embryos using UV-uncaging of caged fluorescein, followed by whole mount immunolabeling to amplify the signal from the uncaged fluorescein.

A protocol to perform lineage tracing and functional genetic analysis of candidate genes at a single cell level using mosaic analysis with double markers (MADM) is presented. MADM clonal analysis provides a quantitative framework to measure the proliferative behavior, cellular output, and lineage relationship of individual progenitors and their daughter cells.
